SkySilence Target Gene Sets are available in a flexible portfolio of lentiviral and retroviral vectors with multiple promoter options including doxycycline-inducible systems along with selectable markers and fluorescent reporters. All shRNAs are designed using the shERWOOD™ algorithm, developed from functional testing of over 250,000 sequences, to deliver potent, single-copy gene knockdown with confidence. The constructs uses an alternate microRNA scaffold called “UltramiR“. The UltramiR scaffold has been optimized for increased shRNA processing and potency based on the key determinants for primary microRNA processing (Auyeung et al 2013).

SkySilence™ shRNA Whole Genome Arrayed Library

Reliable arrayed shRNA for systematic functional genomics

SkySilence™ shRNA arrayed libraries are a great resource for core labs. All shRNA constructs are designed using the shERWOOD algorithm which is based on the functional testing of over 250,000 shRNA sequences using a high-throughput sensor assay (Knott et al 2014) and uses key sequence characteristics for predicting shRNA potency to select the rare shRNA designs that are potent at single copy representation in the genome.  The library uses an alternate microRNA scaffold called “UltramiR“. The UltramiR scaffold has been optimized for increased shRNA processing and potency based on the key determinants for primary microRNA processing (Auyeung et al 2013). Over 18,000 human genes targeted. Each human gene is targeted by an average of 6 shRNA constructs
