Skyang Biotech cDNA collections offer a convenient solution that decreases the time required to start an experiment and increases the chances of success. Creating cDNA clones from biological material requires time, labor and money. While cDNA clone production is a well-established process it requires three major steps: synthesis, cloning and validation.  Rather than spending time on cloning, let us streamline your workflow.

Our CRISPR-Cas9 lentiviral reagents include a range of lentiviral expression vectors expressing guide RNA (gRNA) or gRNA+Cas9. The sgRNA expression vectors are designed to select for high gene-editing efficiency using direct indicators of CRISPR-Cas9 expression from fluorescent reporters or selection markers. Target gene sets are provided as bacterial glycerol stocks or ready-to-use lentiviral particles. The CRoatan* algorithm gRNA designs result in superior knockout efficiency. We offer a wide range lentiviral vector formats and further customization options for dual gRNA constructs, CRISPRa, CRISPRi or Cas9 nickase.

ORF

Skyang Biotech ORF Collections provide a comprehensive resource of fully annotated, sequence-verified entry ORFs derived from collections generated by the Dana Farber Cancer Institute-Center for Cancer Systems Biology (in collaboration with the Broad Institute) and the international ORFeome Collaboration (OC). The ORF clones are provided in Gateway adapted entry vectors for fast and convenient transfer to any compatible expression vector. Open Reading Frame (ORF) clones contain only the protein coding sequence allowing for the addition of fusion tags.

shRNA reagents are next generation vector-based RNAi triggers designed using the proprietary shERWOOD** algorithm developed and validated in Dr. Gregory Hannon’s laboratory at Cold Spring Harbor Laboratory (Knott et al 2014). An alternate microRNA scaffold “UltramiR” has been optimized for increased shRNA processing and potency based on new information on the key determinants for primary microRNA processing (Auyeung et al 2013).

In pooled shRNA screening, hundreds to thousands of hairpins are combined (pooled) and interrogated simultaneously in a multiplex assay without the need for robotics or liquid handling.

Share your target gene list with us and we will design gRNAs/shRNAs targeting each gene. Or if preferred, share your gRNA/shRNA designs for cloning into our lentiviral vector. Each clone is sequence-verified and arrayed into 96-well plates as glycerol stocks, plasmid DNA, or ready-to-use lentiviral particles
